The Defense Advanced Research Projects Agency (DARPA) is seeking innovative proposals under the D-PECHE research opportunity to demonstrate proof-of-concept for new mechanisms that write and erase epigenetic base modifications in DNA. The work focuses on developing novel writer and/or eraser mechanisms, with validation of targeted base modification detection and epigenome editing capabilities in living cells. The effort is a four-month program centered on nucleic acid chemistry, protein engineering, and synthetic biology, and it excludes established DNA methylases and demethylases as well as human or animal subjects research. Submissions are evaluated on scientific merit, relevance to DARPA’s mission, and cost realism. Abstracts are due May 27, 2026 at 4:00 PM ET, followed by invited oral proposal packages, and accelerated awards are limited to costs not exceeding $2,000,000.
